Fungal Identification Kit (Carbon Assimilation)

 

Fungal Identification Kit is a system for the identification of the most frequently encountered yeasts. The species which are possible to be identified with this kit are listed in the identification Table at the end of the package insert.

[Summary]

Yeast-like fungus is widely distributed in nature. It infests in all parts of human body as normal flora. While one's immunity lows down, it will endogenous or prone to opportunistic infections. As to the widely misuse of antibiotics and immunosuppressants in these years and the popularity of organ transplantation and catheter technology, prevalence rate and death rate of opportunistic fungal infection are rapidly rising up. Candida has become the fourth pathogen in blood infection. Rapid identification plays an important role in the fungus’ diagnosis and treatment.

[Principle]

Fungal Identification strip consists of 22 cupules containing dehydrated substrates which enable the performance of 21 assimilation tests. The cupules are inoculated with a semi-solid minimal medium and yeasts will grow only in the carbon source which they can utilize.  

Observe the turbbidity change of each well after 24 hours and identify Fungi species referring to the Analytical Index or using the identification software.

[Materials required but not provided]

1) Sabouraud's culture medium

2) General equipments of Microbial laboratory

[Storage and Period of Validity]  12 months under 2~8°C

[Warnings and Precautions]

1)  This kit is for professional in-vitro diagnostic or microbial QC use only.

2) This kit is for one time use only.

3)  Instructions must be followed to obtain accurate results.

4)  Do not use the kit when it’s expired.

5)  All patient samples should be treated as if capable of transmitting diseases.  

[Sample collection]

The kits can not be directly used for identification of clinical specimens. Specimens should be separated in appropriate medium (such as Sabouraud's culture medium) before separation strain identification.  

 [Operation Procedures]

1. Take the culture media and culture plate out of refrigerator, warm them up to the room temperature;

2. Label the sample and note dowm the details;

3. Be sure the pathogenic fungal colonies have been prepared by use of Sabouraud’s Medium or other fungi culture medium, use sterilized loops to take 1~3 colonies, make fungal suspension by adding it to a fungal culture medium tube to get the turbidity to comply with No.2 McB turbidity tube

4. Use an aseptic pipette tip to apply 150μl of fungal suspension to each well including negative and positive control well;

5. Add a drop of aseptic paraffin oil to each well and screw the lid on

6. Place the boards in the incubator and keep incubating for 20~24 hours at 30°C(strains from deep infection incubated at 37°C)

7. Observe the result according to the analytical concordance list or identification software.

 [Result Observation]   

find what strain it is from the analytical concordance list according to the assimilation features.

Usually, the test result can be reported when the positive well changes turbid transparently after 20 to 24 hours. To some strains with lower activity you can extend your observation time properly. It is unavailable to report result when the turbidity of positive well is close to negative well and identification must do again after low down the concentration.   

[Limitations of the kit]

1) Only be used to identify species of fungi in the database;

2) Only be used for the cultivation of a single microorganism colonies.

[Disposal of the discarded]  Thorough incineration.
